Nux Vomica Tincture contains not less than 0.15 per cent w/w and not more than 0.30 per cent w/w for the sum of brucine (C23H26N2O4) and strychnine (C21H22N2O2), of which 43 per cent to 67 per cent is strychnine.
It may be prepared as follows.
Nux Vomica, in No. 710 powder 1 part
Ethanol (70 Per Cent) 10 parts
Mix the powdered Nux Vomica with ethanol (70 per cent) as prescribed. Macerate for at least 10 days with sufficient shaking. Separate the residue from the ethanol and strain under pressure if necessary. Allow the combined liquid to stand for 48 hours and filter.
Dose 0.6 to 2.4 mL.
Other relevant information Taking more medication than recommended doses of nux vomica tincture may produce convulsions of all voluntary muscles due to strychnine toxicity.

Packaging and storage Nux Vomica Tincture shall be kept in tightly closed containers, protected from light.
Identification Complies with the test for Identification C described under Nux Vomica.
Ethanol content 60 to 70 per cent v/v (Method II, Appendix 6.5).
Assay Carry out the determination as described in the “Liquid Chromatography” (Appendix 3.5).
Standard preparation, Mobile phase, Chromatographic System, System Suitability, and Procedure Proceed as directed in the Assay under Nux Vomica.
Assay preparation Transfer about 2 g of Nux Vomica Tincture, accurately weighed, to a 20-mL volumetric flask, add sufficient amount of ethanol (60 per cent), mix, and dilute with the same solvent to volume.
Calculation Calculate the contents of brucine and strychnine in the portion of the Tincture taken by the expression:

in which Au is the area of the peak due to brucine or to strychnine obtained from Assay preparation; As is the area of the peak due to brucine or to strychnine obtained from Standard preparation; w is the weight, in g, of Nux Vomica Tincture in Assay preparation, W is the weight, in g, of Brucine RS or of Strychnine RS in Standard preparation; P is the declared content of C23H26N2O4 in Brucine RS, or C21H22N2O2 in Strychnine RS.
Other requirements Complies with the requirements described under “Tinctures” (Appendix 1.16).
